學(xué)術(shù)研究:
劉吉開(kāi)1988年獲得博士學(xué)位,1995年任中山大學(xué)化學(xué)系教授。2002年獲國(guó)家杰出青年基金。2008年成為“973”首席科學(xué)家。研究成果曾獲2003年度國(guó)家自然科學(xué)二等獎(jiǎng)、CAS-Bayer獎(jiǎng)、2007年度云南省自然科學(xué)一等獎(jiǎng)、2009年獲談家楨生命科學(xué)創(chuàng)新獎(jiǎng)。2006年獲中國(guó)科學(xué)院優(yōu)秀教師稱(chēng)號(hào)(指導(dǎo)研究生)。至今,以第一作者或通訊作者在國(guó)內(nèi)外發(fā)表學(xué)術(shù)論文300多篇,其中SCI論文200多篇,影響因子10以上的有8篇,影響因子5以上的有24篇。著作1部,獲國(guó)內(nèi)外授權(quán)專(zhuān)利10項(xiàng)(包括3個(gè)國(guó)際專(zhuān)利授權(quán))。他現(xiàn)擔(dān)任國(guó)際期刊Nat. Prod. & Bioprospect. 主編,J. Chem. Ecol.等4個(gè)國(guó)際期刊的編委,以及國(guó)內(nèi)多種期刊編委。曾獲德國(guó)洪堡研究獎(jiǎng)學(xué)金,在德國(guó)留學(xué)多年,在德國(guó)拜耳公司藥物研究中心從事過(guò)藥物研究工作。
系統(tǒng)地開(kāi)展了高等真菌藥物化學(xué)和生物活性成分研究,并形成了特色,所領(lǐng)導(dǎo)的研究團(tuán)隊(duì)目前已成為世界上高等真菌化學(xué)和生物活性研究的中心之一。已完成200多種高等真菌的化學(xué)成分研究,并對(duì)高等真菌子實(shí)體和發(fā)酵液提取物進(jìn)行了多方面的活性研究,分離鑒定了2000余個(gè)不同類(lèi)型的化合物,其中有700多個(gè)新化合物,包括10個(gè)新的結(jié)構(gòu)骨架類(lèi)型,發(fā)現(xiàn)了一系列結(jié)構(gòu)新穎、有價(jià)值的生物活性物質(zhì)。經(jīng)生物活性和藥理學(xué)試驗(yàn),發(fā)現(xiàn)有些化合物具有抗腫瘤、糖和脂代謝調(diào)節(jié)、抗艾滋病毒、致幻等多種顯著的生物活性,有多個(gè)分子目前作為候選藥物分子在評(píng)價(jià)之中。發(fā)現(xiàn)的化合物中先后有10多個(gè)被國(guó)際天然產(chǎn)物權(quán)威雜志Nat. Prod. Rep.(《天然產(chǎn)物報(bào)告》)列為“研究熱點(diǎn)”。從云南新種蘑菇(Trogiavenenata)中發(fā)現(xiàn)了兩個(gè)新的非蛋白質(zhì)氨基酸毒性成分,并證實(shí)誤食該蘑菇是30多年來(lái)導(dǎo)致云南不明猝死的原因(相關(guān)結(jié)果見(jiàn)Angew. Chem. Int. Ed.2012, 51: 2368 ; Science 2010, 329: 132; 2012, 335: 1293)。根據(jù)研究結(jié)果,隨后對(duì)其進(jìn)行干預(yù),印刷了大量宣傳冊(cè),在危險(xiǎn)和猝死發(fā)生地區(qū)進(jìn)行散發(fā),警告人們不要食用該菌。干預(yù)見(jiàn)到了明顯效果,2010以來(lái)沒(méi)有出現(xiàn)類(lèi)似猝死報(bào)告。研究解決了長(zhǎng)期困擾科學(xué)界的難題,挽救了200多個(gè)寶貴的生命!
從韌革菌(Boreostereum vibrans)發(fā)酵液中發(fā)現(xiàn)結(jié)構(gòu)新穎并具有胰脂肪酶抑制活性的天然活性物質(zhì)-韌革菌素(vibranlactone),通過(guò)結(jié)構(gòu)改造與修飾,發(fā)現(xiàn)一比原型化合物活性提高500倍的活性化合物, IC50達(dá)到nM級(jí),與現(xiàn)在市售的陽(yáng)性對(duì)照藥 Xenical(Olistat)相當(dāng)。與有關(guān)人員合作闡明了其生物合成途徑(相關(guān)結(jié)果見(jiàn)專(zhuān)利ZL 200610048625.0; Org. Chem. 2006, 8: 5749; Angew. Chem. Int. Ed. 2013, 52: 2298)。還闡明了韌革菌中結(jié)構(gòu)多樣的雜萜類(lèi)化合物的生物合成機(jī)制;發(fā)現(xiàn)了其中氧化脫羧關(guān)鍵反應(yīng)的單氧化酶VibMO1;證明了由異戊烯基對(duì)羥基苯甲醇通過(guò)發(fā)散生物合成途徑(a divergent pathway)形成韌革菌素類(lèi)雜萜。其中3-取代的γ丁內(nèi)酯類(lèi)化合物Vibralactone G 新穎的生物合成途徑,系首次在天然產(chǎn)物化學(xué)中報(bào)道(Angew. Chem. Int. Ed. 2016, 55: 5463–5466)。現(xiàn)正按1類(lèi)新藥進(jìn)行開(kāi)發(fā)。
研究發(fā)現(xiàn)了一種先前在歐洲一直被當(dāng)作食用菌、不被人們認(rèn)知的危險(xiǎn)毒蘑菇——棕灰口蘑(Tricholoma terreum)。從這種蘑菇的子實(shí)體中共分離得到十五個(gè)結(jié)構(gòu)新穎的三萜,包括terreolides A–F (1–6)和 saponaceolides H–P (8–16)。其中化合物1-3具有獨(dú)特的5/6/7三環(huán)螺環(huán)結(jié)構(gòu),而化合物4-6是新骨架三萜。進(jìn)一步研究發(fā)現(xiàn),化合物7和13,在蘑菇子實(shí)體中含量大,經(jīng)小鼠口服急毒測(cè)試,發(fā)現(xiàn)其半數(shù)致死量分別為88.3和63.7 mg/kg,并且這兩個(gè)化合物都可以引起小鼠體內(nèi)肌酸激酶水平的升高。以上發(fā)現(xiàn)說(shuō)明,棕灰口蘑很可能是10多年前在《新英格蘭醫(yī)學(xué)雜志》(N. Engl. J. Med. 2001, 345: 798)上報(bào)道的橫紋肌溶解死亡病例的重要原因。該項(xiàng)研究結(jié)果發(fā)表在《歐洲化學(xué)》上(Chemistry-A European Journal 2014, 20: 7001-7009; Chemistry-A European Journal 2016, 22: 5786-5788)。
天然來(lái)源的fucosylated glycosaminoglycan(FG)具有非選擇性的內(nèi)源性Tenase抑制活性,由于復(fù)雜多糖的化學(xué)研究存在技術(shù)上的困難,30多年來(lái)其確切結(jié)構(gòu)尚存疑問(wèn);此外因具有F.XII與血小板激活活性,天然FG的實(shí)際應(yīng)用價(jià)值有限。采用糖苷鍵選擇性解聚以及寡糖純化技術(shù)制備了一系列來(lái)源于天然FG的寡糖化合物,并首次通過(guò)純化寡糖化合物的結(jié)構(gòu)解析闡明了復(fù)雜FG的確切化學(xué)結(jié)構(gòu)(側(cè)鏈Fuc取代基的存在方式及其主鏈連接方式);通過(guò)構(gòu)效關(guān)系比較研究,首次報(bào)道了強(qiáng)效抑制內(nèi)源性Tenase系列純化寡糖化合物以及最小寡糖片段--FG九糖(化合物5)。選擇性Tenase抑制劑的發(fā)現(xiàn)不僅有利于進(jìn)一步闡明生理性止血與病理性血栓形成的機(jī)制差異,而且具有重要的新藥研發(fā)價(jià)值。實(shí)驗(yàn)動(dòng)物研究顯示,所述FG九糖具有類(lèi)似依諾肝素(LMWH)的抗血栓活性,在等效抗血栓劑量的高倍給藥劑量下,依諾肝素可顯著增加大鼠出血量,而FG九糖則對(duì)出血傾向無(wú)顯著影響。作為主要合作者研究的該新藥項(xiàng)目已申請(qǐng)一系列國(guó)內(nèi)及國(guó)際(PCT)發(fā)明專(zhuān)利保護(hù), 其成果發(fā)表于《美國(guó)科學(xué)院院刊》(Proc. Natl. Acad. Sci. USA, 2015, 112, 8284-8289)上,并以首期4000萬(wàn)的價(jià)格轉(zhuǎn)讓給企業(yè)。
