學(xué)術(shù)研究
1、民族藥物層級(jí)庫(kù)的構(gòu)建
課題組深入民族地區(qū)(鄂西、新疆、青海、西藏、廣西、貴州、云南)廣泛收集了1000余種中草藥和民族藥物,構(gòu)建了中草藥和民族藥物樣本庫(kù)(含1000種藥材樣本)→藥材提取物庫(kù)(3000個(gè)提取物,一種藥材含石油醚、乙酸乙酯、甲醇三個(gè)提取物)→預(yù)組分化庫(kù)(正在建設(shè))→民族藥物成分庫(kù)(1000個(gè)以上民族藥單體成分)。依據(jù)新藥研發(fā)的經(jīng)典規(guī)律—Lipinski五規(guī)則,我們采用特殊的柱色譜前處理技術(shù)構(gòu)建預(yù)組分化庫(kù)。
2、抗糖尿病藥理活性及機(jī)制研究
長(zhǎng)期從事天然藥物抗糖尿病活性成分篩選與機(jī)制研究工作。利用最新發(fā)展的前沿技術(shù)(基因編輯CRISPR-Cas9、CETSA、靶蛋白垂釣)探索與藥物與靶蛋白間相互作;構(gòu)建基于L6細(xì)胞IRAP-mOrange熒光轉(zhuǎn)染促GLUT4轉(zhuǎn)膜活性篩選方法,實(shí)現(xiàn)藥物抗糖尿病活性快速篩選;已建立多種飲食干預(yù)、基因表達(dá)干預(yù)的糖尿病動(dòng)物、細(xì)胞模型,全面研究藥物作用效果;利用基因工程技術(shù)構(gòu)建基因沉默/過(guò)表達(dá)載體調(diào)控信號(hào)分子表達(dá),實(shí)現(xiàn)藥物作用機(jī)制研究。
3、基于中草藥和民族藥物庫(kù)的抗腫瘤活性化合物的發(fā)現(xiàn)
長(zhǎng)期從事天然藥物抗多種腫瘤活性與機(jī)制研究;團(tuán)隊(duì)已成熟構(gòu)建體內(nèi)外抗腫瘤活性物質(zhì)篩選平臺(tái),實(shí)現(xiàn)組分或單體化合物體抗腫瘤藥效系統(tǒng)評(píng)價(jià);團(tuán)隊(duì)將結(jié)合腫瘤發(fā)病的熱門(mén)靶點(diǎn),探索天然藥物抗腫瘤活性的新機(jī)制;采用MTT和CKK-8方法篩選上述構(gòu)建的中草藥和民族藥物提取物庫(kù),在各種柱色譜分離純化技術(shù)的基礎(chǔ)上結(jié)合活性導(dǎo)向、質(zhì)譜導(dǎo)向以及核磁共振氫譜導(dǎo)向技術(shù),快速、高效、靶向地發(fā)現(xiàn)抗腫瘤活性成分。
3、中藥和民族藥化學(xué)成定性和定量分析及溯源研究
應(yīng)用UPLC-Q-TOF/MS結(jié)合UNIFI數(shù)據(jù)庫(kù)快速自動(dòng)表征中藥和民族藥的化學(xué)成分,并用UPLC-TQ-MS/MS方法對(duì)主要成分進(jìn)行準(zhǔn)確定量。最后通過(guò)結(jié)合生物活性篩選方法確定主要藥效成分;熟練應(yīng)用UPLC-Q-TOF/MS與UPLC-TQ-MS/MS技術(shù),利用非靶向代謝組學(xué)的方法區(qū)分中藥材和民族藥不同品種,明確同一中藥材不同品系在化學(xué)組成上的共性及區(qū)別;基于UPLC-MS與1HNMR等技術(shù),結(jié)合非靶向代謝組學(xué)(PCA、PLS-DA、OPLS-DA)的方法對(duì)中藥和民族藥材進(jìn)行產(chǎn)地溯源,以區(qū)分不同產(chǎn)地的中藥材和民族藥及評(píng)價(jià)其質(zhì)量。
4、中藥和民族藥新劑型研究
進(jìn)行中藥靶向遞藥系統(tǒng),中藥制劑體內(nèi)藥動(dòng)學(xué)及成藥性評(píng)價(jià);中藥新劑型與新制劑的研究開(kāi)發(fā)。在從事中藥及民族藥復(fù)方制劑的開(kāi)發(fā)工作中,其參與企業(yè)研發(fā)的兩個(gè)中藥六類(lèi)新藥“川七鎮(zhèn)腦寧膠囊”和“赤母婦炎寧膠囊”已獲臨床批件。
5、中藥和民族藥新品種選育
主持選育蘄艾新品種蘄特1號(hào),參與選育蘄艾新品種2個(gè)(蘄特2號(hào)和3號(hào))。
